John was born in New Kent County, St. Paul's Parish. Some people claim he was born about 1703, but he owned property by 1719, which implies he was at least 21 by that time. He was probably born about 1695. He was the son of Thomas Poindexter and Sarah Crafford. His father died when he was very young and his mother soon married Rev. James Brechen, a prominent Anglican minister. John was probably schooled at home by his step-father, as it is apparent from later events that he had a very good education for that period. About 1727, he married Christian Gissage, the daughter of Richard Gissage and Sarah Shelton.
John began to aquire land besides that which he had inherited from his father. In 1738, he bought 400 acres in Hanover County, adjoining his own land, and on the same day, he patented an additional 400 acres. In 1739, he bought another 1000 acres in the same county. Finally, in 1752, he patented 400 acres in Louisa County. This land might have been all together, for the tracts in Hanover County might have been in that part of the county that became Louisa County in 1742.
When Louisa County was created, a County Court was established, and Justices were appointed for life by the Royal Governor. John was one of the original Justices appointed. John is also listed in the records of the Fredericksville Parish as a vestryman. John made his will in 1750, in which he mentions his wife and all his children. John's mother, Sarah Brechin, died in 1752 and left land to John which she had inherited from her father. John died between November 1752 and January 1753, his will was proved on May 22, 1753 in Louisa County. His wife Christian died in 1779. Ann Poindexter, who married Reuben Slaughter, was a daughter in this family.
